Systemic lupus erythematosus (SLE) is a chronic, inflammatory autoimmune disorder . It may affect the skin, joints, kidneys, and other organs.

Causes
SLE (lupus) is an autoimmune disease. This means there is a problem with the body's normal immune system response. Normally, the immune system helps protect the body from harmful substances. But in patients with an autoimmune disease, the immune system can't tell the difference between harmful substances and healthy ones. The result is an overactive immune response that attacks otherwise healthy cells and tissue. This leads to chronic (long-term) inflammation. The underlying cause of autoimmune diseases is not fully known. Some researchers think autoimmune diseases occur after infection with an organism that looks like certain proteins in the body. The proteins are later mistaken for the organism and wrongly targeted for attack by the body's immune system.

What is the Burden of Systemic Lupus Erythematosus (lupus) in the United States? Systemic lupus erythematosus affects a conservatively estimated 322,000 to possibly over a million people in the United States. [Due to the lack of definitive epidemiological information, the exact number of people with lupus is currently unknown.] About nine out of 10 people who have lupus are women. Approximately one third of deaths occur among men and women younger than 45. During 1979-1998, the annual number of deaths from lupus rose from 879 to 1,406 and the crude death rate increased from 39 to 52 per million population, with a total of 22,861 deaths reported during this 20-year period. Systemic lupus erythematosus (SLE) can affect persons of any age, but strikes mostly young women of childbearing age. Lupus causes the immune system to attack its own body cells. Lupus can cause severe joint and muscle pain, extreme exhaustion, fevers, skin rashes, and can lead to organ failure and death.

Significance And Limitations Of New Lupus Gene Expression Research
ScienceDaily (Jan. 21, 2008) See also: Mary K.Crow, M.D., associate chief of the division of Rheumatology and director of Rheumatology research at Hospital for Special Surgery in New York, has written an editorial in The New England Journal of Medicine to accompany the papers. One study appears in The New England Journal of Medicine and the other three appear in Nature Genetics, all to be published online on Jan. 20. In her editorial, Dr. Crow talks about the importance of the studies and the questions they leave unanswered, along with insights on next steps in lupus research. "Overall, these papers confirm what investigators have been finding over the past decades," says Dr. Crow, the co-director of the Mary Kirkland Center for Lupus Research and director of the Autoimmunity and Inflammation Research Program at Hospital for Special Surgery. "They show that many aspects of the immune system are involved in the development of the disease, but they also provide a new level of detail regarding the specific molecular pathways that contribute." The new research looks at how variations in a single DNA base pair, called single nucleotide polymorphisms (SNP), can be used to identify genetic variations among individuals that are associated with a diagnosis of lupus. In some cases, the SNPs call attention to important genes but have no apparent functional importance themselves. In other cases, the variations might actually change the protein product so that it functions differently in lupus patients and healthy individuals. For example, in one specific gene the frequency of an "A" nucleotide in people with lupus might be significantly greater than in healthy individuals without the disease. This small variation may alter the expression or function of the protein encoded by that gene in a way that contributes to the disease.
