A neurodevelopmental disorder that affects females, Rett syndrome is associated with cortical atrophy, stereotypical hand movements and severe mental deficiency. With an incidence of 1 in 10,000 - 15,000, it is one of the most common causes of mental retardation in females. Rett syndrome is characterized by loss of acquired skills after a period of normal development in infancy. Mutations in X-linked methyl-CpG binding protein 2 (MECP2) which cause Rett syndrome include both nonsense (R168X and R255X) and missense (R106W and R306C) mutations. Genetic analysis for Rett Syndrome at the Greenwood Genetic Center includes complete sequencing of the entire coding region of the MECP2 gene. Further testing by MLPA for deletions and duplications is available for individuals that test negative via sequencing, but continue to carry a clinical diagnosis of Rett syndrome. Specimen requirements : 5 to 10 ml of peripheral blood collected in an EDTA (lavender top) Vacutainer tube is preferred. The minimal blood needed for reliable DNA isolation is 3 ml. If necessary, ACD solution A Vacutainer tubes (yellow top) may be submitted. Please contact the laboratory for more information.

(Redirected from Rett syndrome Jump to: navigation search Introduction This syndrome is rare and occurs only 1 in 15,000 to 20,000 girls aged 1 to 18 years (based on studies in Sweden, the US and Japan). Although this is a genetic abnormality, less than 1% of the cases are inherited conventionally as most occur spontaneously in the last stages of pregnancy. It is confined almost exclusively to girls. There is a mutation in the MECP2 gene on the X chromosome , but this in not necessarily immediately obvious. Course Of Illness It takes usually until the age of 6 to 18 months to express itself by various symptoms such as problems crawling or walking. There is a progressive loss of purposeful use of her hands and the ability to speak. There are 4 stages that have been defined in the more severe expression of the disease (see more details under Ref. 2 below. Some of the symptoms are repetitive hand movements such as moving the hands repeatedly to the mouth or clapping, wringing, washing motions. The muscles are showing weakness causing gait problems. Some features are similar to autism, but it is a different neurological entity. Diagnosis A child neurologist needs to be involved in the diagnosis of this condition. The specialist will go over the symptoms in detail and rule out other conditions. A genetic test will help to diagnose about 70% to 80% of the cases (MECP2 gene assay), but 20% to 30% will be due to other genetic abnormalities. At this point there is no reliable therapy for this rare condition. More research is needed to possibly develop a genetic vaccine in the future.

What follows is from reader Ingrid Fossgard-Moser, whose daughter Kaia (the raving beauty in the picture) has Rett Syndrome, a rare neurological disorder. You can find out more about it here . Big thanks to Ingrid for sending this in. "My daughter is different. Any mother will tell you this but my daughter is beautiful. Kaia has eyes you will never forget and she looks like an angel. She is the perfect cuddle, keeps any secrets, has a quirky sense of humour and prefers water to air and maybe music to speech. Kaia also has Rett syndrome.
Rett is a severe and rare neurological condition that affects girls almost exclusively with a prevalence rate around one in every 15000 to 20000. These girls suffer from profound and multiple handicaps and always remain dependent on others for help.
Kaia turned two in May but is unlike other two-year-olds. She can sit independently (although regularly topples over) but has never crawled or walked so still has a baby’s body rather than one of a typical toddler transformed by constant activity. Girls with Rett syndrome rarely have any speech (and if they do they do they almost always loose this by the age of three) so we are no longer waiting for words but are immensely proud that Kaia has recently learnt to blink her eyes for Yes.
There are no toddler tantrums and terrible twos to try our patience and no sibling rivalry and bickering with her nearly four-year-old brother that friends complain about. Instead, she adores him like a best friend and Tage has a special tenderness reserved for his sister – perhaps helped by an ability to opt in and out of playing with her with no danger of her wrecking his toys or pulling his hair. He understands her better than anybody, says she always agrees with him and reassures me when I struggle to get her attention: “Mummy, Kaia is just thinking.”

A girl is born with RETT SYNDROME every 5 hours, but most people have never heard of it. It is a severe, crippling disorder that affects baby girls who develop normally until 6-18 months of life, when they begin to lose acquired skills. By the age of three years, girls with Rett syndrome face severe lifelong disabilities. Due to severe apraxia, they have a will and a desire to move, but have difficulty directing their movements or speech. Curvature of the spine, seizures, irregular breathing, difficulty chewing, and swallowing problems are some of the many challenges faced by girls with Rett syndrome. Psych.; MAPS Rett syndrome is a disorder occurring in girls who, prior to the onset of the disorder (usually between six and eighteen months) have appeared to develop normally (Perry, Sarlo-McGarvey, & Haddad, 1991. Rett syndrome is characterised by a course of development involving sequential progression through a four-stage clinical grouping. As the child progresses through the stages the phenotype changes - with each stage having defined characteristics, and the onset of new behaviours or symptoms and the reduction of others (Sansom, Krishnan, Corbett, & Kerr, 1993). The disabling and handicapping effects of the syndrome are multifaceted with symptoms of the syndrome impacting on cognitive as well as physical development. Rett syndrome has been described as leaving children "profoundly mentally and physically disabled" (Saunders, McCulloch, & Kerr, 1995 p. 496).
Attempts to explain the pathophysiology of Rett syndrome have raised several possibilities but no conclusive evidence. Several early studies pointed to abnormal dopamine levels (Van Acker, 1991). Lloyd et al, (1981, cited in Van Acker) found some characteristics of Rett syndrome were also characteristic of boys with Lesch-Nyhan, a syndrome where dopamine levels have been shown to have a causal relationship with the behaviour related (Van Acker, 1991). Recent studies however have failed to replicate this finding (Van Acker, 1991).
