This causes the cells to become stuck in capillaries which deprives the body of oxygen and causes inflammation . The disease usually occurs in periodic painful attacks, eventually leading to damage of some internal organs, stroke , or anemia , and usually resulting in early death.
Treatment
The first approved drug for the treatment of sickle cell anaemia, hydroxyurea[?] , was shown to decrease the number and severeness of attacks in a study in 1995 and shown to increase survival time in a study in 2003. Hydroxyurea had previously been used as a cancer drug, and there is some concern that long-term use may be harmful, but it is likely that the benefits outweigh the risks.

The gene allele responsible for sickle cell anaemia is incompletely recessive . A person who receives the defective gene from both father and mother develops the disease; a person who receives one defective and one healthy allele remains healthy, but can pass on the disease and is known as a

Font Font The red blood cell becomes sickle shaped instead of round shaped usually resulting from genetics as a person inherits one abnormal gene from a parent. Sickle cell anemia is not contractible the only way to obtain the disease is by inheriting the abnormal cells. A normal blood cell that contains the oxygen carrying component hemoglobin is shaped like a doughnut. In case of Sickle cell anemia, the hemoglobin becomes abnormal causing blood cells to stack together producing a sticky collection of cells that blocks oxygen to organs and tissues. These cells clog the veins and arteries not allowing blood to flow properly. The oxygen in the blood is hampered and cannot supply enough to all tissues and organs in the body to keep it healthy.

Incomplete dominance means that the heterozygote's phenotype is mixed, somewhere between the two homozygote phenotypes. So what does 'incompletely recessive' mean? The most accurate description of sickle cell disease's genetics is recessive gene#Sickle cell anemia (although I would regard sickling as co-dominant, because cells in the heterozygote appear normal under most conditions but sickling occurs with hypoxaemia). In view of the lack of response, I have changed the articles myself. Axl 13:27, 31 Jan 2005 (UTC) I'm afraid I can't answer these questions, however I'd like to take the welcome opportunity to make a few suggestions, questions:
  • "baseline anemia" what is this? Can this terminology be explained? "Reticulocyte counts are elevated, reflecting new red blood cells replacing the rapidly cleared older cells - red blood cell life span is markedly reduced in this disease." - what does "Reticulocyte counts" mean? "Vasoocclusive crises" - why are they called this? Where does the term come from?

Cause
The sickle cell syndromes are caused by a mutation in the b-globin gene that changes the sixth amino acid from glutamic acid to valine. HbS (a2b26Glu®Va1) polymerizes reversibly when deoxygenated to form a gelatinous network of fibrous polymers that stiffen the erythrocyte membrane, increase viscosity, and cause dehydration due to potassium leakage and calcium influx. The prototype disease, sickle cell anemia, is the homozygous state for HbS. In this condition more than 80% of the red cell hemoglobin is HbS. In the deoxygenated state HbS moelcules link to form chains and these result in sickling and increased rigidity of the red cells. Sickling produces a shortening of red cell survival and an obstruction of the microcirculation.

  • inherited blood disorder affecting the oxygen carrying part of the blood cells called hemoglobin hemoglobin in sickle cell anemia forms a "sickle" or banana shape
    • normal hemoglobin is called hemoglobin A sickle cell hemoglobin is called hemoglobin S
    this shape causes it to be broken down in the liver leading to not enough blood cells people with sickle cell disease experience pain, damage to organs, and sometimes early death
    • this is due to sickle shaped blood cells blocking tiny blood vessels this leads to prevention of blood flow to certain areas of the body
    edit Diagnosis
    • diagnosis is made by a blood test called hemoglobin electrophoresis
      • those with mostly hemoglobin S have sickle cell disease those with only hemoglobin A do not have sickle cell disease those with both hemoglobin A and hemoglobin S are heterozygote carriers and have sickle cell trait
        • this means they are carriers of the sickle cell gene they generally do not have any health problems but can pass the gene to their children
        this test requires 3cc of blood in a red-capped EDTA tube
