Human Genome Project
An international project to map all human genetic material The aim of the Human Genome Project, which was launched in 1990 by an international team of researchers, was to decode all 3 billion base pairs in the human genome. In 1995 Germany joined the team and went on to characterise the base sequence of around 1.5 per cent of the genome. In 2001 it was announced that every human gene had been mapped. Scientists discovered that only three to five per cent of the DNA is present in the form of genes. The remaining DNA consists of regulatory elements, deactivated pseudogenes and regions of repeating sequences.

        • a. chop up DNA, ligate to DNA cloning vector to form recombinant molecule, insert into host cell, replicate/amplify… b. cloning vector must:
          • 1. be capable of autonomous replication in host cell 2. must have a restriction site 3. must have a gene that you can select for (ie:antibiotic resistance)
          c. first vectors with prokaryotic plasmids which are still widely used d. development of methods using bacteriophage (23kb size of avg human gene), cosmids (44kb) and then finally YAC’s and BAC’s (1Mb) allowed for the HGP to be successful

Also by John Buchel: Related Stories: by John Buchel Wednesday, April 16, 2003 The International Human Genome Sequencing Consortium announced Monday that it had completed the mapping of the human genome two years ahead of schedule and $300 million under its originally projected budget. The success of what has come to be known as the Human Genome Project is expected to change the way human DNA is examined and interacted with. In June 2000, the consortium announced it had finished a rough draft of the sequence and had since then worked to polish its decoding techniques. The current vision of the genome has been sequenced to an accuracy of 99.99 percent and is considered highly reliable. The next and most immediate task for the international consortium will be dissemination of its findings to the rest of the scientific community.

The $3 billion project was founded in by the United States Department of Energy and the U.S. National Institutes of Health , and was expected to take 15 years. Due to widespread international cooperation and advances in the field of genomics (especially in sequence analysis ), as well as huge advances in computing technology, a rough draft of the genome was finished in 2000 (announced jointly by US president Bill Clinton and British Prime Minister Tony Blair on June 26 ), two years earlier than planned. The consortium comprised:
Initiation of the Project was the culmination of several years of work supported by the US Department of Energy, in particular a feasibility workshop in 1986 and a subsequent detailed description of the Human Genome Initiative in a report that led to the formal sanctioning of the initiative by the Department of Energy. This 1987 report stated boldly, "The ultimate goal of this initiative is to understand the human genome" and "Knowledge of the human genome is as necessary to the continuing progress of medicine and other health sciences as knowledge of human anatomy has been for the present state of medicine". Though leaders had already been considering technologies required for such an undertaking at least as early as 1985
