Nel 1890, insieme al tedesco Robert Koch e al giapponese Kitasato Shibasaburo , scopr¬ che, iniettando siero prelevato da un animale affetto da tetano in un secondo animale, quest’ultimo diveniva immune alla malattia; il siero dell’animale immunizzato poteva, quindi, essere iniettato in un terzo individuo, con il risultato che anch’esso sviluppava immunit  alla stessa malattia. Con il contributo del batteriologo tedesco Paul Ehrlich , Behring estese quindi le sue ricerche, applicandole con grande successo alla prevenzione della difterite infantile.

Nachdem frühzeitig seine schulische Begabung erkannt wurde, ermöglichten ihm verschiedene Stipendien die Ablegung des Abiturs; das von ihm angestrebte Studium der Medizin schien ohne weitergehende Unterstützung jedoch nicht finanzierbar, weshalb Behring die militärärztliche Laufbahn einschlug, in der für jedes Studiensemester ein Dienstjahr angerechnet wurde. Am 2. Oktober 1874 trat er in die Kaiser-Wilhelm-Akademie für das militärärztliche Wesen in Berlin ein. Zugleich schloss er sich dem damals dort ansässigen Corps Suevo-Borussia - einer Studentenverbindung - an. 1878 wurde er promoviert, 1889 an das Institut für Infektionskrankheiten in Berlin zu Robert Koch berufen, wo er im Sommer 1890 die Grundlagen für die Serumtherapie entwickelte und in Gemeinschaft mit dem Japaner Shibasaburo Kitasato das Tetanusantitoxin entdeckte; seit 1891 arbeitete er - zum Teil unterstützt durch Paul Ehrlich - an der Entwicklung der Diphtherieantitoxine, letztendlich ermöglichte dies den Sieg über diese - bis dahin oftmals tödlich verlaufende - Krankheit. 1895 übernahm Behring den Lehrstuhl und die Leitung des Hygienischen Instituts der Universität Marburg, wo 1914 auch die Behring-Werke entstanden. 1917 erlag er in seinem Marburger Wohnhaus einer Lungenentzündung.
