Prace g³ównie z syntezy polipeptydów. Za opracowanie prostej i szybkiej, zautomatyzowanej laboratoryjnej metody syntezy ³añcuchów polipeptydowych (w du¿ych ilo¶ciach, na tzw. sta³ej matrycy) otrzyma³ w 1984 Nagrodê Nobla w dziedzinie chemii. Metoda Merrifielda jest obecnie stosowana zarówno w laboratoriach, jak i zak³adach przemys³owych.

Parcours universitaire N©   Fort Worth (Texas), Robert B. Merrifield effectue des ©tudes de chimie   l’universit© de Californie   Los Angeles (UCLA). Jeune dipl´m©, il est engag© par un laboratoire de biochimie. Il s’aper§oit tr¨s vite de ses limites dans ce domaine et d©cide de retourner   l’universit© pour approfondir le sujet. Il obtient son doctorat de biochimie en 1949, et rejoint aussit´t l’Institut de recherche m©dicale Rockefeller (actuellement universit© Rockefeller)   New York, o¹ il effectue toute sa carri¨re (de postdoctorant   professeur titulaire en 1966). Travaux scientifiques Les travaux de Robert B. Merrifield concernent la synth¨se des prot©ines. Celles-ci sont des cha®nes d’acides amin©s connect©s entre eux par des liaisons peptidiques. Les peptides ont la mªme structure que les prot©ines, mais leurs cha®nes d’acides amin©s sont plus courtes. Partant de leurs constituants chimiques, on peut cr©er des peptides, puis fabriquer des prot©ines enti¨res. Avant que Robert B. Merrifield ne mette au point une nouvelle m©thode de synth¨se dans les ann©es 1950, ce processus ©tait fastidieux et tr¨s long. Pour que les acides amin©s s’associent dans le bon ordre, il fallait bloquer certaines parties, afin que seuls les sites voulus interagissent et se connectent. Entre chaque ©tape, le peptide devait ªtre isol© et purifi©. Lorsqu’on ©liminait les sous-produits de la r©action pr©c©dente, on perdait irr©m©diablement une partie de la structure du peptide.

Robert Bruce Merrifield wurde am 15. Juli 1921 in Fort Worth (Texas) geboren. Im Jahr 1966 wurde er zum Professor an die Rockefeller University in New York berufen. Merrifield f¼hrte Forschungen zur chemischen Synthese an festen Matrizen durch. Dabei entwickelte er u. a. ein schnelles Verfahren zur Synthese von Proteinen und Nucleins¤uren , das nach ihm benannte Merrifield-Verfahren. Svilupp² un metodo per la sintesi chimica su supporto solido delle proteine che diminu¬ drasticamente il tempo necessario a compiere questo processo (da alcuni mesi a pochi giorni), permettendo ai ricercatori di incrementare lo sviluppo di anticorpi sintetici e di vaccini. Per il suo contributo alla sintesi delle proteine, Merrifield fu insignito del premio Nobel per la chimica nel 1984. Merrifield si laure² in chimica all’Universit  della California a Los Angeles nel 1949. Nello stesso anno si trasfer¬ al Rockefeller Institute for Medical Research (ora Rockefeller University) a New York, dove rimase per tutta la carriera. Le proteine sono costituite da catene di amminoacidi tenuti uniti da legami peptidici. I peptidi hanno la stessa struttura delle proteine, ma sono costituiti da catene di amminoacidi pi¹ brevi. Gli scienziati sintetizzano artificialmente peptidi per produrre intere proteine. Prima che Merrifield, alla fine degli anni Cinquanta, sviluppasse la nuova tecnica, il processo era lungo e tedioso. Per ottenere che gli amminoacidi si legassero nella corretta sequenza, certe parti dovevano essere rese inattive in modo che solamente i siti desiderati potessero interagire e legarsi. Tra un passaggio e l’altro, il peptide doveva venire isolato e purificato. Quando i sottoprodotti della reazione precedente venivano eliminati, ineluttabilmente parte della struttura del peptide andava persa.
