Si occupò delle variazioni d'intensità delle radiazioni cosmiche con altezza sul livello del mare e dell'intensità della radiazione cosmica nella profondità dei laghi montani. Effettuò la prima determinazione fotoelettrica dell'energia dei quanti di luce e studiò il campo del limite delle radiazioni ultraviolette. L'esperimento che lo rese famoso però fu quello relativo alla misura della carica elettrica, che effettuò immobilizzando tramite un campo elettrico piccolissime gocce d'olio che si formavano nell'aria ionizzata da raggi X: è conosciuto col nome di 'esperienza di Millikan'.
