Eric Martz wrote: 1ALX seems a bona-fide example of altLocs, but is unusual in having four, or even five, altLocs for some atoms. It also has sequence microheterogeneity, marked by insertion codes in column 27, and described in the remarks. The one thing that seems inconsistent is that each of the four altLocs A,B,C,D for MOH519 (methanol) have 50% occupancy, totalling 200%. This I don't understand. MOH519 has five altLocs A,B,C,D,E each with 40% occupancy. 1ALX displays just fine in the 10.x. Basically it is a set of solvent molcule options around double-stranded DNA. One can select each different option using conformation 0;wireframe on or select them one by one with conformation 1;wireframe on conformation 2;wireframe on conformation 3;wireframe on etc. There are always PDB files that are illegally formatted. Already I see such cases for altLoc, thanks to Jaim's list below. The most common case appears to

History
The first electron microscope was built in by Ernst Ruska . It was greatly developed through the and has allowed great advances in the natural sciences . The advantage of an electron beam is that it has a much smaller wavelength (see wave-particle duality ), which allows a higher resolution - the measure of how close together two things can be before they are seen as one. Light microscopes allow a resolution of about 0.2 micrometres , whereas electron microscopes can resolutions below 1 nanometer
Process
Electron beams from a cathode are focused by magnetic lenses on to the specimen. They are then magnified by a series of magnetic lenses until they hit photographic plate or light sensitive sensors - which transfer the image to a computer screen. The image produced is called an electron micrograph (EM).
Types
The Transmission electron microscope (TEM) produces 2D images (for example of cells ) while the Scanning electron microscope (SEM) produces 3D images or models. As its name implies the TEM image is produced by detecting electrons that are transmitted through the sample. By contrast the SEM usually monitors secondary electrons which are emitted from the surface due to excitation by the primary electron beam. Generally, the TEM resolution is about an order of magnitude better than the SEM resolution, however, because the SEM image relies on surface processes rather than transmission it is able to image thicker samples and gives better 3D contrast.

Instead of using normal (visible) light, an electron Microscope uses electrons. With this type of microscope we see details smaller then 10nm. Another advantage is the large field of depth compared with an optical microscope. A Scanning Electron Microscope (SEM) looks at the surface, with a Transmission Electron Microscope (TEM) you can see through a specimen. A Focused Ion Beam (FIB) is able to carefully remove the surrounding of an interesting detail of a specimen. Combined with the right accessory the Electron Microscope can give detailed information about the elemental composition, crystal structure etc.

    G.Völksch) In order to characterize the microstructure of glasses and glass-ceramics, electron optic imaging, i.e. methods based on the interaction of electron beams with the respective materials are necessary. In the Otto-Schott-Institut, transmission electron microscopy (TEM) also of replicas and with the aid of electron diffraction as well as electron microprobe analysis is used. Scanning electron microscopy (SEM) including energy dispersive (EDX) and wavelength dispersive x-ray analysis (WDX) and cathodoluminescence imaging (CL) is carried out. The traditional imaging of fractured surfaces using replica techniques and TEM is predominantly used for all types of phase separated samples. Here, the direct imaging of ion etched samples is hardly possible because of small or negligible contrast. Using replica techniques does not allow using EDX and therefore conclusions on the chemical composition are not possible. The main use of TEM, however, is imaging and sample characterizations by EDX and electron diffraction at precipitated micro or nano crystals. Amain topic is the characterization of precipitated crystallites in order to produce nano particles in the systems B O -Na O-Al O -In O -SnO and B O -CaO-Na O-Fe O In-situ O )in a SEM was carried out in corporation with the Institut für Materialforschung, Bayreuth University (Dr. H.-J. Kleebe).
