    Electron Microscopy
    The function of the Central Facility for Electron Microscopy is to provide hands-on access to electron microscopes, sample preparation equipment plus data collection and data reduction instrumentation, along with advice, training and research collaboration. The scope of the Facility is materials characterization of the topography, morphology, ele-mental composition, crystalline microstructure, crystal defects, and atomic arrangements of materials, largely on a scale from 10 micrometers down to the near-atomic level.

Electron Microscopy
The mission of our laboratory is to aid the clinical and research community in their ultrastructural investigations, whether it is for research or diagnostics, using either transmission electron microscopy (T.E.M.) or scanning electron microscopy (S.E.M.). We offer a full range of services including, consultation, sample preparation, training on instrumentation or documentation of ultrastructure by our staff. We are prepared to carry out all of the preparation and electron microscopy or share it with the investigator or the investigators designate. Recharge rates supplied upon request.

    Summary
    At its meeting in April 1998, the Cell Biology and Biophysics (CBB) Subcommittee of the National Advisory General Medical Sciences (NAGMS) Council identified a need for better methods for examining large macromolecular assemblies and for imaging macromolecules in vivo . Biological structure on these scales is not accessible to analysis by X-ray crystallography or NMR, but is approachable by high-resolution electron microscopy (EM). This workshop was convened to evaluate prospects for utilizing EM to (1) determine the atomic resolution structures of isolated macromolecular assemblies in solution and (2) determine 3-D macromolecular arrangements in cells in conjunction with the known atomic structures of isolated components. Session 1 examined the hierarchies of biological structure and the capabilities and complementarily of existing methods. Session 2 assessed current capabilities and limitations of EM for structure determination of macromolecules, macromolecular assemblies, and cells.

Principles of electron microscopy
Before we can understand how the electron microscope (EM) works, it is important that we distinguish between resolution and magnification. Magnification is how much bigger we can make something appear. Resolution is how much better we can distinguish two closely spaced points. A lower limit on resolution is imposed by the fact that when we view specimens in a microscope, we are viewing them using waves. We use light waves to view objects under the light microscope. It is vitally important to understand that we cannot distinguish details finer than the wavelength of these waves. This is because light diffracts (bends into what 'should' be crisp shadows) and interferes with itself when moving around very small objects (or through very small holes). This means edges become fuzzy, and images lose their crispness.

Equipment We are well-equipped for electron microscopy studies through the research of Professor David Cherns Professor John Steeds and Dr Roger Vincent and their groups. Currently, we have four transmission electron microscopes (TEMs) and two scanning electron microscopes (SEMs): we have a range of equipment for sample preparation, including mechanical and chemical polishing equipment, dimplers, ion thinners and access to a focussed ion beam thinner. JEOL 2010 TEM, acquired by the Group in 2006 Convergent beam electron diffraction pattern (with acknowledgement to Dr B Krause) Imaging, diffraction and microanalysis We do imaging, diffraction (in particular convergent beam electron diffraction) and microanalytical studies. Analytical facilities include energy dispersive X-ray microanalysis (EDX), electron energy loss spectroscopy (EELS) and cathodoluminescence (CL).
