New Scientist reported the combination of two notorious killer viruses, HIV and Ebola , in an attempt to find an effective gene therapy vector for the treatment of cystic fibrosis. When this work was presented at a scientific meeting the audience laughed out loud. Gene therapy is targeted at virtually every ill known to human beings, especially those inhabiting the first world, including pain relief, cosmetic hair replacement and muscle building. Massive investment has gone in but no clinical efficacy has ever been proven, despite anecdotal claims of success. Last year in the US, gene therapy clinical trials ground to a halt amid scandalous reports of deaths and conflicts of interest

Arlo Miller arlo@thebiotechclub.org The completion of the human genome project has brought us one step closer to understanding the molecular basis of health and disease. The removal of this bottleneck from the drug discovery process shifts the emphasis toward translating this knowledge into clinical practice, specifically, validating specific genes as determinants for disease. However, for many important diseases, genetic determinants were identified many years ago. Unfortunately, knowledge of the genetic mutation has not led to effective drugs in many cases. While the standard approach to most diseases is to formulate a drug that can modulate the symptoms or to supplement missing proteins, gene therapy represents a more intuitive approach of directly correcting or replacing aberrant or missing genes. This approach carries the hope of a permanent cure. While progress in gene therapy has been difficult, its potential benefits are so overwhelming that many companies are working in this area and have gene therapy drugs in trials. Diseases Targeted Over 50% of gene therapy trials are for the treatments of various cancers such as head and neck, ovarian, or metastatic cancers. In these cases the idea is to deliver a toxic gene to the cancer cells or to deliver an immunogen to stimulate specific immune responses that destroy the cells. Most of the remaining trials address traditional single gene genetic disorders such as ADA deficiency, hemophilias, cystic fibrosis, and hemoglobin disorders.

Gene Therapy
In gene therapy, a type of virus known as a retrovirus is used to replace a defective gene in unhealthy cells with a new, healthy gene. Scientists insert the healthy gene into the ribonucleic acid (RNA) of the retroviruses. These retroviruses are mixed with unhealthy cells taken from a patient and cultured in a laboratory. The retroviruses infect the unhealthy cells with the healthy gene, adding the healthy gene to the deoxyribonucleic acid (DNA) of the cells. The work of pioneer researchers show convincing evidence that genetic changes are being discovered in patients where vaccine reactions appear to be casually involved. Thus, it is also plausible that genetic changes could be taking place as a result of the current childhood vaccination programs. This could be affecting many of our children nationally, here in the US, and globally. This could also be what's affecting the thousands of Gulf War Veterans with Gulf War Syndrome (GWS). Yet another burden of proof to be solved: Vaccines are not causing adverse genetics. Text Size Gene Therapy - Putting Muscle Into the Research Bio-artificial muscle implants could attenuate or perhaps prevent muscle loss among astronauts in orbit and patients with muscle-wasting diseases on Earth. When astronauts spend time in space, they also spend some of the hard-earned muscle mass that they've built up on Earth prior to their flights. In the microgravity environment of a spacecraft in orbit, muscles atrophy quickly. The body perceives that it does not need muscles to be as strong, resulting in a decrease of muscle mass by as much as 5 percent a week - up to about 20 percent over time in the muscles that are used more to fight gravity on Earth, such as those in the calves and along the spine.

Gene Therapy Oversold by Scientists Who Disregard Risks
Angela Ryan reports on the continuing fiasco of attempts to genetic engineer human beings. New Scientist reported the combination of two notorious killer viruses, HIV and Ebola [1], in an attempt to find an effective gene therapy vector for the treatment of cystic fibrosis. When this work was presented at a scientific meeting the audience laughed out loud. Gene therapy is targeted at virtually every ill known to human beings, especially those inhabiting the first world, including pain relief, cosmetic hair replacement and muscle building. Massive investment has gone in but no clinical efficacy has ever been proven, despite anecdotal claims of success. Last year in the US, gene therapy clinical trials ground to a halt amid scandalous reports of deaths and conflicts of interest [2]. The US National Institutes of Health (NIH) set up a special telephone hot line for victims that counted 652 cases of serious adverse events along with six unexplained deaths. Effects included high fevers, infections and severe changes in blood pressure, all of which went previously unreported to the NIH Recombinant DNA Advisory Committee (RAC). David Baltimore, Nobel laureate and president of Caltech, a gene therapy based biotech company, said " I disagree we've had any benefit from gene therapy trials so far, many of us are now asking, what the hell are we doing putting these things into people?"
