Restriction enzymes are proteins that cut deoxyribonucleic acid (DNA) chains. DNA makes up the units of heredity called genes and as such, forms the blueprint for all life. Arber's work allowed researchers to modify the DNA molecule and paved the way for important advances in biotechnology, such as the manufacture of synthetic hormones (proteins that control the body's functioning) for medical use. Arber shared the Nobel Prize with American biologists Hamilton O. Smith and Daniel Nathans, who contributed to the discovery of restriction enzymes.

Au d©but des ann©es soixante, Arber d©couvre le syst¨me qui permet   certaines bact©ries de rendre inoffensif un bact©riophage (virus parasite des bact©ries). Il montre que la bact©rie produit une enzyme dite « de restriction  » qui reconna®t l’ADN du bact©riophage intrus et le d©coupe, ce qui le neutralise. Au cours de ce processus, la bact©rie produit aussi une autre enzyme, qui empªche l’enzyme de restriction de d©truire son propre ADN. Arber ©met aussi l’hypoth¨se que ces enzymes de restriction reconnaissent des sites pr©cis de la s©quence d’ADN, et ne la coupent qu’en ces endroits. Cette hypoth¨se est confirm©e par le biochimiste am©ricain Hamilton O. Smith, qui d©montre que l’enzyme d©couverte par Arber d©coupe effectivement l’ADN en des sites pr©cis. Il d©couvre ©galement d’autres enzymes de restriction bact©riennes, qui ont la propri©t© de d©couper chacune l’ADN en des endroits diff©rents. Enfin, Daniel Nathans, un coll¨gue de Smith, trouve une application   ces enzymes : il les utilise pour ©tudier le g©nome d’un virus du singe (le SV 40), qu’il d©coupe en divers morceaux enfin d’en ©tablir une sorte de carte. Aujourd’hui, les enzymes de restriction repr©sentent un outil essentiel pour le g©nie g©n©tique.
