Zernike invented the phase-contrast, or phase, microscope, a microscope that can show tiny differences in the way a transparent specimen bends light ( see Optics ). The phase-contrast microscope is especially useful in studying living tissue. For that invention, Zernike was awarded the 1953 Nobel Prize in physics. The son of two mathematics teachers, Zernike was born in Amsterdam, the Netherlands. He earned his Ph.D. degree in physics in 1915, at the University of Amsterdam. In 1913 he accepted a position as assistant to the Dutch astronomer Jacobus Kapteyn at the University of Groningen, where Zernike became a lecturer in theoretical physics two years later. In 1920 he was promoted to full professor of theoretical physics, and in 1941 expanded his chair to include mathematical and technical physics and theoretical mechanics. Later in his career, Zernike was briefly affiliated with Johns Hopkins University in Baltimore, Maryland.

V roce 1913 se stal asistentem na universitì v Groningenu a setrval tam potom ještì v letech 1920 až 1958 jako profesor. Jeho døívìjší práce v optice se zamìøovala pøedevším na astronomické dalekohledy. Když zkoumal vady optických pøístrojù, pøišel na princip fázovì-kontrastního mikroskopu. Proslavil se tím, že byl schopen rozlišovat svìtelné paprsky procházející skrz prùhledný materiál. V roce 1938 postavil mikroskop, který byl na tomto principu založen. V roce 1952 byl odmìnìn Rumfordovou medailí, kterou pøedávala Královská spoleènost v Londýnì.

Pour cela, il invente un microscope qui utilise un diaphragme pour diriger et focaliser la lumi¨re sur le sp©cimen ©tudi© et une plaque de diffraction, ou plaque de phase, plac©e entre les deux lentilles qui composent l’objectif. Entre la lumi¨re directe et la lumi¨re diffract©e se cr©e une diff©rence de chemin optique telle que leur superposition donne, par interf©rence constructive, une intensit© lumineuse renforc©e. Ce microscope permet d’observer des d©tails indiscernables   l’aide d’un microscope conventionnel, notamment sur des sp©cimens vivants pour lesquels l’emploi indispensable d’un colorant est fatal.
